Current Rroject: Detailed comparative genomics to investigate genome evolution of the newly sequence Brachyspira hyodysenteriae and Brachyspira pilosicoli

Project Description: Whole genomic sequence data from the pathogenic intestinal spirochaetes Brachyspira hyodysenteriae and Brachyspira pilosicoli will be subjected to extensive comparative genomic analysis to help identify potential virulence factors for these two species. Analysis will include comparisons with other species of pathogenic bacteria. Genes theoretically identified as having potential to contribute to virulence will be assessed for their distribution across the genus. These will then be cloned, and recombinant products produced and further characterised. Gene inactivation studies will be carried out, and Brachyspira strains with mutations induced in these genes will be tested for their ability to contribute to bacterial colonisation and induction of disease. Gene products will be tested for their immunogenicity and capacity to induce protective immunity in animals, and hence as vaccine components.
